Applications: The line was produced from a fusion of P3/NSI/1-Ag4-1 cells with malignant cells from a human nodular lymphoma. 
The cells are deficient in hypoxanthine phosphoribosyltransferase (HPRT) and are excellent fusion partners for human B cells. 
Originally the cells secreted human IgM with a lambda light chain; however, with continuous passage they spontaneously lost the ability to secrete immunoglobulin. 
Tested and found negative for ectromelia virus (mousepox).

Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Temperature: 37.0°C 
Atmosphere: air, 95%; carbon dioxide (CO2), 5%
Subculturing: Protocol: Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 2 X 10(5) cells/ml and maintain between 1 X 10(5) and 1 X 10(6) cells/ml. 
Medium Renewal: Every 2 to 3 days
Preservation: Freeze medium: complete growth medium, 95%; DMSO, 5% 
Storage temperature: liquid nitrogen vapor phase
