This is a hypertriploid human cell line having the modal chromosome number of 77 occurring in 46% of cells. Cells with higher ploidies were found at 16.6%. The karyotype of this cell line is very complex. Between 45% and 55% of chromosomes in each cell had structural aberrations, including: der(4)t(1;4) (p13.1;q35); t(15q22q); del(2) (p13); der(7)t(7;?) (p22;?) [7p+]; t(7q22q?) and others. The t(15q22q), del(2), and t(7q22q?) (and several others) had two copies in most cells. There were two types of der(7): one was the 7p+ described above; the other, 7p++, was distinctly longer than N1. There were more than ten markers that were present in some cells only. One DM-like chromosome was found in most cells. Normal N7 and N21 were absent. One normal X and one or two normal Y chromosomes were detected.
In vitro transformation properties were assayed at various early passages. Hs 746T exhibited gross alteration in size, shape and staining of cells with bizarre nuclear and nucleolar shapes. Colonies formed in Methocel and on confluent monolayers of normal human epithelial cells and fibroblasts.
Propagation:
ATCC complete growth medium: The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%. Temperature: 37.0°C
Subculturing:
Protocol: Remove medium, and rinse with 0.25% trypsin, 0.03% EDTA solution. Remove the solution and add an additional 1 to 2 ml of trypsin-EDTA solution. Allow the flask to sit at room temperature (or at 37C) until the cells detach.Add fresh culture medium, aspirate and dispense into new culture flasks. Subcultivation Ratio: A subcultivation ratio of 1:2 to 1:8 is recommended Medium Renewal: 2 to 3 times per week
